Age-Dependent Changes in Organ-Specific Oxidative Damages and Glutathione Redox Cycling Efficiency in Rats

Abstract
We examined organ specific differences in oxidative damages and glutathione system as a function of rat age. Oxidative damages in the liver, brain, testis, kidney and heart were monitored by the level of lipid peroxidation and protein carbonyl content. As one of defense parameters against oxidative damages, tissue specific activities of glutathione utilizing and redox cycling enzymes in rats were analyzed. Our results indicated that the degree of protein carbonylation showed better correlation with age than that of lipid peroxidation. The redox cycling efficiency of glutathione transferase activity was rather increased in most of organs except testis up to old age. These age-related biochemical changes were organ-specific rather than universal. In addition, the drastic biochemical changes at a specific point of aging led us to assume that there might be a certain threshold in aging process, In order to understand th mechanism of aging, it is therefore recommended that research should be concentrated on biological change occurring during critical transitional period of the aging process.
